In the opening scene of the pilot episode, a young guy named Joey hits on Lorelai.

The humour is that Joey obviously has no idea who Jack Kerouac is. (This probably ruined whatever slim chances he might have had with Lorelai.)
Jack Kerouac is best known for his novel On The Road, based on his travels across America. Thus, Lorelai’s summons of the Kerouac name in response to Joey’s comment, “I’ve never been here before. Just passing through…”
