Rory makes this statement in reference to the massive amount of lipgloss and other makeup that Lorelai carries in her purse.
RuPaul is a famous drag queen from the 90’s. (S)he gained fame by doing things like recording with Elton John (“Don’t Go Breakin’ My Heart”) and, after being hired by MAC Cosmetics, became the first drag supermodel. Hence the makeup reference.
